ProMutor is platform for generating advantageous point mutants based on explicit modelling of evolutionary history.

Here, the effect of suggested mutations is evaluated by predicting their outcomes based on explicit modelling of the evolutionary history of natural sequences. The resulting multiple-sequence-alignment-based conservation scores \(\Delta \Delta E\) quantify how likely each of the 19 substitutions are in terms of what has been observed in the evolutionary record, with \(\Delta \Delta E \approx 0\) corresponding to very conservative substitutions (have minor effect on protein structure) and the lower \(\Delta \Delta E\) is, the more likely those mutations disrupt protein's structure. Note, that one should compare scores between mutants with different numbers of introduced mutations. In addition, scores should not be interpreted as a quantitative measure of protein's properties (e.g. stability). They are used to rank mutants and select the most advantageous ones (those with the highest evolutionary score among others).
